冻存和复苏的原则:慢冻快融》当细胞冷到零度以下,可以产生以下变化:细胞器脱水,细胞中可溶性物质浓度升GAO,并在细胞内形成冰晶。如果缓慢冷冻,可使细胞逐步脱水,细胞内不致产生大的冰晶;相反,结晶就大,大结晶会造成细胞膜、纲胞器的损伤和破裂。复苏过程应快融,目的是防止小冰晶形成大冰晶,即冰晶的重结晶。慢冻程序》1.标准程序:采用细胞冻存器》当温度在-25℃以上时,1~2℃/min;当温度达-25℃以下时,5~10℃/min;当温度达-100℃时,可迅速放入中。2.简易程序:将冷冻管(管口要朝上)放入纱布袋内,纱布袋系以线绳,通过线绳将纱布袋固定于罐罐口,按每分钟温度下降1~2℃的速度,在40min内降至表面过夜,次晨投人中。3.传统程序:冷冻管置于4℃10分钟→-20℃30分钟→-80℃16~18小时(或隔夜)→槽长期储存。细胞冻存方法:1.预先配制冻存》(1)8%DMSO+细胞生长(92%血清)2.取对数生长期细胞,经胰酶消化后,加入适量冻存,用吸管吹打制成细胞悬(1×10(6)~5×10(6)细胞/ml)。3.加入1ml细胞于冻存管中,密封后标记冷冻细胞名称和冷冻日期。长期保存。保存细胞的复苏方法:1.快速解冻》冻存细胞从中取出后,立即放入37℃水浴中,轻轻摇动冷冻管,使其在1分钟内全部融化(不要超过3分钟)。2.解冻后的细胞可直接接种到含完全生长培养的细胞培养瓶中直接进行培养,24小时后再用新鲜完全培养替换旧培养,以去除DMSO。3.如果细胞对冷冻保护剂别敏感解冻后的细胞应先通过离心去除冷冻保护剂,然后再接种到含完全生长培养的培养瓶中。

细胞冻存基本方法:1.细胞:选对数生长期细胞,收集细胞24小时前换一次。2.计数:按常规方法把细胞制成细胞悬,计数,令细胞密度达5×10(6)/ml,离心去上清,吸入离心管中。3.冻存:先用培养9分+DMSO 1分(或甘油)配成10%的DMSO冻存,按上法一滴滴加入离心管中,然后用吸管轻轻吹打令细胞重悬。4.分装:分装入无菌安剖中,每安剖加1.5毫升细胞悬。5.封口:用塑料安剖时拧紧瓶口即可;如用火焰封闭安剖口后,仔细检查,定要封严,必要时可浸入蓝色中观察,为安全起见,把安剖缝入纱布小袋中,以防浸入后,融解时因受热发生爆炸伤人;纱袋一端系以线绳,末端扎有小牌,注明:细胞名称,冻存日期,以便日后查找。细胞株(系)的使用,为医学研究和测试工作带来了大的方便。但细胞的传代是有限制的,长期连续传代的细胞,不仅消耗大量的人力和物力,而且细胞的生长与形态等会有一定退变或转化,因而细胞失去原有的遗传性,有时还会由于细胞污染而造成传代中断,种子丢失。因此,在实际工作中常需冻存一定数量的细胞,以备替换使用。细胞冷冻与复苏是细胞培养 室的常规工作和通用技术。

细胞培养无菌操作的演示:凡是带入超净工作台内的酒精、PBS、培养基、胰蛋白酶的瓶子均要用75%酒精擦拭瓶子的外表面;靠近酒精灯火焰操作。器皿使用前必须过火灭菌,继续使用的器皿(如瓶盖、滴管)要放在GAO处,使用时仍要过火。各种操作要靠近酒精灯,动作要轻、准确,不能乱碰。如吸管不能碰到废缸。吸取两种以上的使用时要注意更换吸管,防止交叉污染。玻璃器械洗消:一、新的玻璃器皿的洗消:自来水刷洗,除去灰尘。烘干、泡:烤箱中烘干,然后再浸入5%稀中12小时以除去脏物、铅、等物。刷洗、烘干:12小时后立即用自来水冲洗,再用洗涤剂刷洗,自来水冲干净后用烤箱烘干;泡、清洗:用清洁浸泡12小时,然后从缸内捞出器皿用自来水冲洗15次,Zui后蒸馏水冲洗3-5次和用双蒸水过3次。烘干、包装:洗干净后先烘干,然后用牛皮纸(油光纸)包装。GAO压消毒:包装HAO的器皿装入GAO压锅内盖HAO盖子,打开开关和安全阀,当蒸气成直线上升时,关闭安全阀,当指针指向15磅时,维持20-30分钟。GAO压消毒后烘干。二、旧的玻璃器皿的洗消:刷洗、烘干:使用过的玻璃器皿可直接泡入来苏尔或洗涤剂溶中,泡过来苏尔溶(洗涤剂)的器皿要用清水刷洗干净,然后烘干。泡、清洗:烘干后泡入清洁(),12小时后从缸内捞出器皿立即用自来水冲洗(避免蛋白质干涸后粘附于玻璃上难以清洗),再用蒸馏水冲洗3次。烘干、包装:洗干净的器皿烘干后取出用牛皮纸(油光纸)等包装。
