慢病毒感染快速指南
1. 将目的细胞按 30%汇合度接种到 24 孔板，约 3-5×104细胞数/孔，不同细胞因为细胞大小不同细胞数量会不同，快速指南以细胞数量为 5×104为例，加入 500uL 完全培养基进行细胞培养。
2. 接种后 12-20h 感染阴性对照慢病毒（接种细胞至病毒感染细胞时间不宜太长，否则细胞增殖影响细胞密度不利用后期抗性筛选）。
3.加病毒量计算方法：（细胞数×MOI 值/病毒滴度）×103=病毒量（uL），加病毒量见下表。同时加入 Polybrene，每孔加 0.25uL，浓度为 10mg/mL Polybrene，细胞培养液中终浓度为5ug/mL。
4.感染 16-20h 后更换培养基，弃去培养基，每孔加入 500uL 新鲜的培养基。
5.感染后 48-96h 显微镜观察荧光。（注：如果 48h-96h 期间细胞汇合度达到 100%，请及时传代培养）。
6.建议将 48 孔板细胞进行传代，传代 12 孔板，后继续观察荧光，根据细胞状态和荧光细胞比例综合判断细胞感染最佳 MOI 值。一般选择细胞生长良好，感染效率 80%左右的感染条件作为最佳感染条件。（注：100%荧光感染不一定为细胞最佳 MOI 值，因为可能会造成细胞慢病毒感染拷贝数太高，影响细胞状态。
