Exonuclease III( 核酸外切酶III) 是一种特异的双链DNA 3` → 5`核酸外切酶。该酶催化从缺口、平端、凹端及少于4 个碱基的3`- 突出端的3`-OH 处将单核苷酸逐个去除的反应，产生5`- 磷酸核苷。Exonuclease III 具有内在的3`- 磷酸酶活性，因此也可从3`- 磷酸末端降解DNA。另外，该酶还具有类脱嘌呤核酸内切酶活性及核糖核酸酶H 活性。Exonuclease III 与S1 核酸酶联用，可从DNA 片段的末端单向删除序列。
