背景资料：CD46 acts as a cofactor for complement factor I, a serine protease which protects autologous cells against complement-mediated injury by cleaving C3b and C4b deposited on host tissue. It may be involved in the fusion of the spermatozoa with the oocyte during fertilization. CD46 acts as a co-stimulatory factor for T-cells which induces the differentiation of CD4+ into T-regulatory 1 cells. T-regulatory 1 cells suppress immune responses by secreting interleukin-10, and therefore are thought to prevent autoimmunity. A number of viral and bacterial pathogens seem to exploit this property and directly induce an immunosuppressive phenotype in T-cells by binding to CD46.

我现在做elisa，目的是利用此法来检测我样品中蛋白的浓度，我用的是改良的双抗体夹心法，就是包被鼠单抗，加标准品和待测样品，再加兔多抗，再加山羊抗兔的酶标二抗，TMB显色。我现在的问题是摸索单抗和多抗的稀释度，所以就将多抗稀释了三个浓度1:5000,1:10000,1:15000,1:20000，单抗 包被稀释度是1:15000，标准品都是五倍稀释即：25μg/mL , 5μg/mL , 1μg/mL , 0.2μg/mL , 0.04μg/mL 酶标二抗是1:5000 双波长A450
