The fluorescence intensity is highest between the ratio of F/P=4-12 between cyanine dyes and biomolecules. If the F/P value is too high, the fluorescent probe will self quench and affect the biological activity of biomolecules. It is best to label biomolecules with monosuccinimide ester, but crosslinking was not found with double modified CyDye NHS. The F/P of CyDye NHS labeled antibodies can reach 5-6 at pH (8.5-9.4) for 10 minutes, while there is almost no reaction at pH 7.0. We labeled anti glutathione-S transfer (GST) polyclonal antibodies with different proportions of Cy3 and found that the F/P values obtained when labeled with 1:1, 5:1, 10:1, and 20:1 were 0.28:1, 1.16:1, 2.3:1, and 4.6:1, respectively.
