The PLPro (Papain-Like Protease) substrate is a small 5-mer fluorogenic peptide (Z- RLRGG-AMC) used to measure the activity of SARS family PLPro proteases. Release of the C-terminal AMC dye (7-amido-4-methylcoumarin ) by proteolysis generates fluorescence, which can be monitored at an excitation wavelength of 345 nm and an emission wavelength of 445 nm.Conditions optimized for use with: Papain-like Protease (SARS-CoV-2) Assay Kit: Protease Activity (BPS Bioscience #79995). The peptide corresponds to a short sequence of ubiquitin and may also be used as substrate of USP5 (isopeptidase T) and ubiquitin C-terminal hydrolases (UCH family, for example UCHL1).
