描述：DNA 错配修复系统（MMR）主要由三种蛋白组成，包括 MutS、MutL 和 MutH。其中 MutS 负责识别错配或未结合位点并与之结合，随后 MutL 蛋白与 MutS-DNA 形成复合体，增强其稳定性，并激活 MutH 的内切酶活性，协同解旋酶和单链结合蛋白将错配序列切除。 ttMutS 蛋白是一种分离自嗜热栖热菌（Thermus thermophilus）的DNA 错配修复蛋白，在最适温度 65-75℃ 范围内，该蛋白具有依赖于 DNA 的 ATPase 活性。 Tth MutS 蛋白在DNA 复制中识别错配碱基和插入碱基形成的 Loop 结构，其紧密结合 Loop 结构从而阻止聚合酶的延伸。因此，其可用于错配 PCR 反应（ARMS-PCR）和基因芯片中，除此外，在基因合成中能阻止错配的产生，降低基因突变率。
