    尽管JC-1在许多实验室中被广泛使用，但其水溶性差会带来极大的不便。即使在1μM浓度下，JC-1也倾向于在水性缓冲液中沉淀。JC-10被开发为需要高染料浓度的JC-1的优越替代品。与JC-1相比，JC-10具有更好的水溶性。JC-10能够选择性地进入线粒体，并随着膜电位的增加，其颜色从绿色可逆地变为橙色。该特性是由于膜偏振时JC-10聚集体的可逆形成，导致发射光从520nm（即JC-10单体形式的发射）到570nm（即J-聚集体的发射）的偏移。当在490nm处激发时，随着线粒体膜变得更加偏振，JC-10的颜色从绿色可逆地变为绿橙色。使用通常安装在所有流式细胞仪中的滤光片可以检测这两种颜色，因此可以在荧光通道1（FL1）中分析绿色发射，在通道2（FL2）中分析绿橙色发射。除了在流式细胞术中的潜在用途外，它还可用于荧光成像和荧光微孔板平台。该试剂盒通过优化的测定方法为所有基本组件提供检测线粒体膜电位丧失的细胞凋亡。该荧光测定基于通过阳离子亲脂性JC-10染料检测细胞中线粒体膜电位变化。在正常细胞中，JC-10浓缩在线粒体基质中，形成红色荧光聚集体。然而，在凋亡和坏死细胞中，JC-10以单体形式存在，并以绿色荧光染色细胞。该试剂盒经过优化，可通过流式细胞术筛选细胞凋亡激活剂和抑制剂。我们还提供方便的 96 孔和 384 孔荧光微量滴定板格式试剂盒 （cat#22800），用于高通量筛选。

    协议摘要
     
    1. 用密度为 5 × 10 的测试化合物制备细胞5至 1 x 106细胞/毫升
    2. 将细胞重悬于500μL的JC-10工作溶液（2-5×105细胞/管）
    3. 在 37°C 或室温下孵育 15-60 分钟
    4. 使用流式细胞仪使用 FL1 通道（绿色荧光单体信号）和 FL2 通道（橙色荧光聚集信号）进行分析

    重要说明
    在开始实验之前，在室温下解冻所有试剂盒成分。

    工作溶液的制备
    将 25 μL 200X JC-10（组分 A）加入 5 mL 检测缓冲液 A（组分 B）中，充分混合制成 JC-10 工作溶液。避光。


    实验方案样本
    1. 用测试化合物处理细胞一段时间以诱导细胞凋亡。设置并行对照实验。

    对于阴性对照：仅用载体处理细胞。

    对于阳性对照：在2 oC，10%CO 37培养箱中用5-2μM的FCCP或CCCP处理细胞15至30分钟。注意：CCCP或FCCP可以与JC-10工作溶液同时添加。可能需要滴定CCCP或FCCP，以获得最佳单个细胞系的结果。
     
    1. 离心细胞得到 2 - 5 × 105每管细胞数。注意：对于贴壁细胞，轻轻提起细胞0.5mM EDTA以保持细胞摄入量，并在用JC-10工作溶液孵育之前用含血清的培养基洗涤细胞一次。
     
    1. 将细胞重悬于500μL JC-10工作溶液中。
     
    1. 将细胞在室温下或在 37 oC、5% CO 2 培养箱中孵育 15 - 60 分钟，避光。注意：适当的孵育时间取决于所使用的单个细胞类型和细胞浓度。优化每个实验的孵育时间。
     
    1. 使用具有FL1通道的流式细胞仪监测荧光强度，以获取绿色荧光单体信号（在凋亡细胞中），FL2通道用于橙色荧光聚集信号（在健康细胞中）。对细胞进行门控，不包括碎片。建议使用FCCP或CCCP处理的细胞进行补偿校正。

    在BD FACS卡利布尔系统流式细胞仪上分析JC-10的典型流式细胞仪设置如下：
    由于细胞类型和培养条件以及单个仪器的差异，建议的初始条件可能需要修改。
    FL1 PMT 电压 366 FL2 PMT 电压 430
    补偿： FL1 – 47.2%
    FL2;FL2 – 47.0% FL1
