重要说明
使用前在室温下解冻所有试剂盒组件。

工作溶液的制备
将 20 μL 硝酸™近红外储备溶液（组分 A）加入 10 mL 检测缓冲液 I（组分 B）中并充分混合。工作溶液在室温下可稳定保存至少2小时。注意：20 μL 硝基近™红外储备溶液足以用于一块板。避光保存。

实验方案样本

1. 为了刺激内源性NO，在细胞培养基或所需的缓冲液（如PBS或HHBS）中用10 μL 10X测试化合物（96孔板）或5 μL 5X测试化合物（384孔板）处理细胞。对于对照孔（未处理的细胞），加入相应量的培养基或复合缓冲液。注意：在添加化合物之前不需要清洗细胞。但是，如果测试化合物对血清敏感，则可以在添加化合物之前吸走生长培养基和血清因子。抽吸后加入 90 μL/孔（96 孔板）和 20 μL/孔（384 孔板）的 1X Hank 盐溶液和 20 mM Hepes 缓冲液 （HHBS） 或您选择的缓冲液。或者，细胞可以在无血清培养基中生长。

 

1. 在细胞板中加入 100 μL/孔（96 孔板）或 25 μL/孔（384 孔板）的硝酸™ NIR 工作溶液。将细胞与测试化合物和硝酸™近红外工作溶液在 37°C 下共孵育所需的时间，避光。注意：请勿去除测试化合物。注意：对于NONOate阳性对照处理：将细胞与硝酸™ NIR工作溶液在37°C下孵育30分钟。除去工作溶液，并将细胞进一步与1mM DEA / NONOate在37°C下孵育30分钟以产生一氧*氮。有关详细信息，请参见图 1。我们使用 Raw 264.7 细胞在 0°C 的细胞培养基中与 5.20X 硝基™近红外、1 μg/mL 脂多糖 （LPS） 和 37 mM L-精氨酸 （L-Arg） 一起孵育 16 小时。有关详细信息，请参见图 2。

 

1. 去除每个孔中的溶液。添加检测缓冲液II（组分C），100孔板为96 μL/孔，25孔板为384 μL/孔。注意：添加检测缓冲液II前不要洗涤细胞。

 

1. 使用酶标仪在Ex / Em = 650 / 680nm（截止= 665nm）下以底部读取模式监测荧光增加，或使用带有Cy5®滤光片的荧光显微镜拍摄图像。
