协议摘要
 

1. 准备细胞 （0.5 - 1×106细胞/毫升）
2. 加入 1 μL 500X 氮化红™
3. 用测试化合物和硝酸™红在37ºC下孵育细胞所需的时间
4. 使用610/20 nm滤光片用流式细胞仪分析细胞

重要事项
在开始实验之前，在室温下解冻所有组分。

储备溶液的制备
除非另有说明，否则所有未使用的储备溶液应分成一次性等分试样，并在制备后储存在-20°C。避免反复冻融循环。

非酸阳性对照治疗储备溶液（50 mM）
将 200 uL ddH 2 O 加入 NONOate 阳性对照（组分 B）的小瓶中，制成 50 mM NONOacte 阳性对照治疗储备溶液。

工作溶液的制备
用测定缓冲液（组分C）稀释50 mM NONOate阳性对照处理储备溶液，制成1-2 mM NONOate阳性对照工作溶液。

实验方案样本

1. 对于每个样品，在您选择的 0.5 mL 温热培养基或缓冲液中制备细胞，密度为 5×105至 1×106细胞/毫升。注意：应单独评估每个细胞系，以确定NO诱导的最佳细胞密度。

 

1. 将 1 μL 500X 亚硝酸™红（组分 A）加入 0.5 mL 细胞悬液中。注意：对于贴壁细胞，用0.5mM EDTA轻轻提起细胞以保持细胞完整，并在用硝酸™红孵育之前用含血清的培养基洗涤细胞一次。

 

1. 将细胞与测试化合物和硝酸™红在37ºC下孵育所需的时间，以产生内源性或外源性NO。 注意：适当的孵育时间取决于所使用的单个细胞类型和测试化合物。优化每个实验的孵育时间。注意：我们使用 Raw 264.7 细胞在 20 ºC 的细胞培养基中与硝酸™红工作溶液、1 μg/mL 脂多糖 （LPS） 和 37 mM L-精氨酸 （L-Arg） 一起孵育 16 小时。

 

1. 将用硝基™红预孵育30分钟的细胞旋转下来。用1mM DEA NOate阳性对照工作溶液重悬细胞，并在37ºC下再孵育30分钟。有关详细信息，请参见图 1。

 

1. 使用流式细胞仪中的610/20 nm滤光片监测荧光强度。对感兴趣的细胞进行门控，不包括碎片。
