培养了快一个月的细胞了，今天终于鉴定出是我想要的血管平滑肌细胞。我养的是大鼠胸主动脉血管平滑肌细胞,采用的是组织贴块法培养。10天后传第一代（图1），16天后第二代（图2），22天后第三代（图3），并做a－actin免疫细胞化学鉴定（图4）。 图1图2图3 图4典型的血管平滑肌细胞生长"hill and valley"现象。
组织化学染色。（α-SM-actin免疫组织化学鉴定）胰酶消化法培养平滑肌细胞1.1 试剂和动物胰酶购自Sigma公司；α-actin抗体购自中山生物技术有限公司；PBS液各成分均为市售分析纯；新生小牛血清购自HyClone；DMEM为Gibco产品。雄性Wistar大鼠（2只，体重175±25 g）购自中国医学科学院实验动物中心。1.2 胰酶消化法原代培养Wistar大鼠处死，迅速取出胸主动脉，浸泡在含无菌PBS的表面皿中，转移到无菌超净台。在表面皿中漂洗主动脉去除残留的血迹，纵向剪开主动脉，把主动脉铺平
