描述：

Ribavirin (Copegus) is an anti-viral agent indicated for severe RSV infection (individually), hepatitis C infection and other viral infections. In this form ribavirin interferes with RNA metabolism required for viral replication. Ribavirin 5’ mono- di- and tri-phosphates are all inhibitors of certain viral RNA-dependent RNA polymerases which are a feature of anti-sense RNA viruses. Ribavirin 5’-monophosphate inhibits cellular inosine monophosphate dehydrogenase, thereby depleting intracellular pools of GTP. Ribavirin enhances host T-cell-mediated immunity against viral infection through helping to switch the host T-cell phenotype from type 2 to type 1.

体外研究：

Ribavirin significantly reduces the efficiency with which progeny subgenomic replicons transfect new cells in the replicon system, although ribavirin has little effect on levels of HCV replication. Ribavirin increases the mutation frequency of HCV, with the highest rates of mutations being found in the NS5A-encoding region. Ribavirin enhances TH1 while inhibiting T 2 cytokine production by stimulated T cells. Ribavirin shows antiviral activity against a variety of RNA viruses and is used in combination with interferon-alpha to treat hepatitis C virus infection. Ribavirin reduces infectious poliovirus production to as little as 0. 00001% in cell culture. Ribavirin's antiviral activity is exerted directly through lethal mutagenesis of the viral genetic material. Ribavirin markedly reduces viral-induced parameters of macrophage activation at physiologic concentrations (up to 500 mg/mL). Ribavirin inhibits the production of IL-4 by Th2 cells, whereas it does not diminish the production of IFN-gamma in Th1 cells. Ribavirin exhibits antiviral activity against a broad range of both DNA and RNA viruses in vitro. Ribavirin is a cytostatic agent and causes a reduction in synthesis of DNA, RNA and proteins in exposed cells. Ribavirin is thought to induce a switch in T-helper cell phenotype from type 2 to type 1.

体内研究：ALT, AST activities and bilirubin levels are significantly loared by administration of JAT in combination with interferon and ribavirin (p4, livers appear to exhibit some liver protection against CCl4 as evident by the presence of normal hepatic cords, absence of necrosis and lesser fatty infiltration. Groups treated with JAT, Peg-interferon and ribavirin separately or in combination shows reduction in the expression of TGF- β and Bax. In the group treated by triple combination of interferon, ribavirin, and JAT, the expression level of p53 is markedly reduced. Ribavirin capsules (400 mg of ribavirin)-treated Wistar rats show a significant decrease in activin-A and significant increase in follistatin at the serum and liver levels. Ribavirin has strong antiviral activity only when ribavirin is combined with either IFN-α or Peg-IFN-α. Ribavirin (40 mg/kg, p.o.) significantly improves the antiviral efficacy of CM-10-18 in mice. Ribavirin inhibits DENV virus infection in cultured cells, but it is ineffective in reducing viremia in monotherapy.
