纳米抗体是由比利时科学家 Hamers 等 1993 年在《Nature》中首次报道,它是一种天然存在的仅由两条重链组成的特殊抗体,只含有重链可变区(VHH, Variable Domain of Heavy Chain Antibody)和两个常规的 CH2 与 CH3 区,该可变区能单独稳定地在体外存在,是目前已知的可稳定结合目标的最小单位,VHH 晶体宽为 2.5nm,长 4nm,分子量仅为传统完整抗体的 1/10(约 15KD)但依然具有完整的抗原识别能力,因此称之为纳米抗体(Nanobody 或 single domain antibody)

- 有更长的 CDR3 区(纳米抗体 CDR3 通常由 16~24 个氨基酸组成,传统单抗 CDR3 仅有 7~12 个氨基酸),,与抗原特异性结合能力更强
