CXCR4是一种多效趋化因子受体，通过其配体CXCL12调节多种生理过程，包括免疫应答、造血、发育过程和促肿瘤作用。它几乎在所有类型的造血细胞中都有表达。CXCR4轴刺激EGFR、MAPK、PI3K/AKT、Wnt/β-catenin和NF-κB相关信号通路，参与肿瘤的发生、生存、侵袭和归巢。CXCR4/CXCL12信号转导也参与转移。目前，CXCR4已在23种不同类型的肿瘤中被检测到，是肿瘤细胞中最常见表达的趋化因子受体，与患者的预后相关。开发CXCR4拮抗剂，下调癌细胞中CXCR4的表达，对相关疾病的治疗具有重要意义。
