摘要

肿瘤微环境通过动态的双向肿瘤-间质相互作用,积极参与胰腺导管腺癌的发病机制。本研究证明,ATM缺陷的肿瘤上皮细胞以一种基因型特异性的方式重编程肿瘤微环境,从而增强癌症的侵袭性。在基因工程小鼠模型、胰腺星状细胞和癌症相关成纤维细胞共培养系统、单核多组学分析以及人类PDAC模型中,ATM丝氨酸/苏氨酸激酶的肿瘤性缺失驱动CAFs向αSMA+肌成纤维细胞分化,且不依赖于p53状态。这些肌成纤维细胞反过来促进了癌症的侵袭性和化疗耐药性。从机制上讲,ATM缺陷增加了活性氧水平和收缩性信号,从而增强了TGFβ1的分泌。药理学抑制TGFβ逆转了肌成纤维细胞分化,使肿瘤对化疗敏感,并在小鼠和人类ATM缺失模型中抑制了肿瘤进展。这些发现揭示了ATM缺陷肿瘤通过TGFβ信号塑造促癌微环境,并确定了同时靶向肿瘤内在和微环境脆弱性作为一种有前景的精准肿瘤学策略。
