The last decade has seen an extraordinary improvement in fluorescence microscopy instruments. These have been mainly in the development of epi-illumination systems, including dichroic beam splitters, matching excitation and barrier filters, and special objectives (see ref. 1 for background information). Routinely we use a JenaLumar fluorescence microscope (Zeiss, Jena, Germany) equipped with oil immersion wide field 100 objective with an adjustable iris diaphragm. According to our experience, the best documentation is obtained when the photographs are taken on Kodak Tri X-Pan film processed in Kodak developer.
