| Background | Me thylation of lysine residues is a common regulatory posttranslational modification (PTM) that results in the mono-, di-, or tri-m ethylation of lysine at ε-amine groups by protein lysine m ethyltransferases (PKMTs). Two PKMT groups are recognized based on structure and catalytic mechanism: class I me thyltransferases or seven β strand enzymes, and SET domain-containing class V me thyltransferases. Both use the me thyl donor S-adenosyl-L-methionine to me thylate histone and non-histone proteins. Class I m ethyltransferases met hylate amino acids, DNA, and RNA. Six met hyl-lysine-interacting protein families are distinguished based on binding domains: MBT, PHD finger, Tudor, PWWP, WD40 repeat, and chromodomains. Many of these display differential binding preferences based on lysine met hylation state. KDM1 subfamily lysine deme thylases catalyze dem ethylation of mono- and di-me thyl lysines, while 2-oxoglutarate-dependent JmjC (KDM2-7) subfamily enzymes also modify tri-m ethyl lysine residues. Most PKMT substrates are histone proteins and transcription factors, emphasizing the importance of lysine met hylation in regulating chromatin structure and gene expression. Lys9 of histone H3 is mono- or di-me thylated by G9A/GLP and tri-me thylated by SETDB1 to activate transcription. JHDM3A-mediated demet hylation of the same residue creates mono-m ethyl Lys9 and inhibits gene transcription. Tumor suppressor p53 is regulated by meth ylation of at least four sites. p53-mediated transcription is repressed following mono-m ethylation of p53 at Lys370 by SMYD2; di-me thylation at the same residue further inhibits p53 by preventing association with 53BP1. Concomitant di-m ethylation at Lys382 inhibits p53 ubiquitination following DNA damage. Mono-met hylation at Lys382 by SET8 suppresses p53 transcriptional activity, while SET7/9 mono-m ethylation at Lys372 inhibits SMYD2 met hylation at Lys370 and stabilizes the p53 protein. Di-m ethylation at Lys373 by G9A/GLP inhibits p53-mediated apoptosis and correlates with tri-me thylation of histone H3 Lys9 at the p21 promoter. Overexpression of PKMTs is associated with multiple forms of human cancer, which has generated tremendous interest in targeting protein lysine meth yltransferases in drug discovery research. |

Pan Di-M ethyl-lysine Rabbit Polyclonal Antibody -20ºC保存,Western一抗稀释液-20ºC或4ºC保存,一年有效。Western一抗稀释液优先推荐4ºC保存,长期不使用可以考虑-20ºC保存,但冻融可能会导致出现轻微的浑浊和少量不溶物。
注意事项:
如果本抗体用于Western blot (WB)、免疫荧光(IF)、免疫细胞化学(ICC)等实验,请注意回收使用过的稀释抗体。回收的抗体通常至少可以重复使用5-10次。稀释后的抗体,包括已经使用过的稀释抗体,请4℃保存。
回收后重复使用的抗体,使用方法同新鲜稀释的抗体。如果在重复使用过程中发现抗体出现轻微混浊现象,可以10,000g离心1-3分钟,取上清用于后续检测。如果回收的抗体出现明显的絮状物或长霉长菌等情况,则可以考虑废弃该抗体。
提供的Western一抗稀释液也可以用于免疫荧光(IF)、免疫组化(IHC)、免疫细胞化学(ICC)等适当用途。如果希望获得最佳的检测效果,请考虑使用上述检测专用的一抗稀释液。
