Intended use
This immunoassay kit allows for the in vitro quantitative determination of Rat dynamin-2,Dynamin-2 concentrations in serum, Plasma, tissue homogenates and Cell culture supernates and Other biological fluids.
Test principle
The microtiter plate provided in this kit has been pre-coated with an antibody specific to Dynamin-2. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Dynamin-2 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Dynamin-2, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Dynamin-2 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

文献和实验我的题目是鸡Mx基因的克隆，我的策略是先根据该基因家族(dynamin family)的氨基酸保守区设计简并引物克隆出该基因cDNA的部分片段，然后根据该片段序列设计基因特异性引物，利用RACE技术得到cDNA的3‘和5’末端，然后根据末端序列设计引物扩增出该基因全长cDNA. 我用简并引物从鸡cDNA中扩增出一个片段，约250bp, 测序后拿来BLAST时却发现，与之同源性最高的并不是其它的Mx基因，而是人的某一个基因，比对的结果中与该序列同源性较高的有人及鼠Mx基因，但是同源性并不
