在基因功能研究过程中，常常使用的一种方法就是将目的基因转到靶细胞内进行瞬时表达（transient expression），以流式细胞仪检测细胞周期的变化，观察转基因对细胞增殖的影响。由于各种转基因方法的不同，转基因的效率也不同，往往是仅有一部分细胞可以表达目的基因，没有转基因的细胞将影响检测的结果。如何将已经转基因的细胞同未转基因的细胞区分开来进行检测，是一个技术上的难点。常用的一种方法就是将表达绿色荧光蛋白的质粒和目的基因共转染；在共转染时，加入目的基因表达质粒的量远高于表达GFP质粒
