Intended use
This immunoassay kit allows for the in vitro quantitative determination of human ribose-5-phosphate isomerase,RPIA concentrations in serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids.
The ELISA is based on the competitive binding enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to RPIA, During the reaction, RPIA in the sample or standard competes with a fixed amount of biotin-labeled RPIA for sites on a pre-coated Monoclonal antibody specific to RPIA. Excess conjugate and unbound sample or standard are washed from the plate. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of RPIA in the samples is then determined by comparing the O.D. of the samples to the standard curve.

文献和实验品，同时也作为判断结果的对照，因此对照品，特别是阳性对照品的基本组成应尽量与检测标本的组成相一致。以人血清为标本的测定，对照品最好也为人血清，因为正常人血清在各种ELISA模式中可产生不同程度的本底。由于大量正常人血甭较难得到，国外试剂盒中的对照品多以复钙人血浆(recalcified human plasma)为原料，即在血浆中加入钙离子，使其中的纤维蛋白质凝固，除去凝块后所得的液体，其组成与血清相似。阴性对照品须先行检测，确定其中不含待测物质。例如HBsAg检测的阴性对照品中不可含HBsAg，最好抗HBs
酯酶，一般采用对硝基苯磷酸酯(p-nitrophenyl phosphate,p-NPP)作为底 物，可制成片剂，使用方便。产物为黄色的对硝基酚，在405nm波长处有吸收峰。用 NaOH终止酶反应后，黄色可稳定一时间。 AP也有发荧光底物（磷酸4-甲基伞酮），可用于ELISA作荧光测定，敏感度较高于用 显色底物的比色法。 3.4 洗涤液 在板式ELISA中，常用的稀释液为含0.05%吐温20磷酸缓冲盐水。 3.5 酶反应终止液 常用的HRP反应
光光度计测量。用于ELISA的优点为可加宽定量测定的线性范围。 HRP对氢受体的专一性很高，仅作用于H2O2、小分醇的过氧化物和尿素过氧化物(urea peroxide)。H2O2应用最多，但尿素过氧化物为固体，作为试剂较H2O2方便、稳定。试剂盒供应尿素过氧化物片剂，用蒸馏水溶解后，在底物缓冲液中密闭、低温（2～8℃）可稳定1年。3.3.2 AP的底物 AP为磷酸酯酶，一般采用对硝基苯磷酸酯(p-nitrophenyl phosphate,p-NPP)作为底物，可制成片剂，使用方便。产物为黄色的对硝
