Intended use
Human Sepp1 ELISA KIT allows for the in vitro quantitative determination of human selenoprotein p,SeP concentrations in serum, Plasma, tissue homogenates and Cell culture supernates and Other biological fluids.
Test principle
The microtiter plate provided in Human Sepp1 ELISA KIT has been pre-coated with an antibody specific to SeP. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for SeP and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain SeP, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of SeP in the samples is then determined by comparing the O.D. of the samples to the standard curve.

品，同时也作为判断结果的对照，因此对照品，特别是阳性对照品的基本组成应尽量与检测标本的组成相一致。以人血清为标本的测定，对照品最好也为人血清，因为正常人血清在各种ELISA模式中可产生不同程度的本底。由于大量正常人血甭较难得到，国外试剂盒中的对照品多以复钙人血浆(recalcified human plasma)为原料，即在血浆中加入钙离子，使其中的纤维蛋白质凝固，除去凝块后所得的液体，其组成与血清相似。阴性对照品须先行检测，确定其中不含待测物质。例如HBsAg检测的阴性对照品中不可含HBsAg，最好抗HBs
