Comments:
Animals were immunized with bovine mesenteric lymph node cells. Spleen cells were fused with NS1 myeloma cells. The antibody is specific for a 90000 dalton molecule expressed on the surface of bovine B cells. This antibody was included in the panel of the International Workshop on Bovine, Sheep and Goat Leukocyte Differentiation Antigens in 1989 and 1992.
ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%. Atmosphere: air, 95%; carbon dioxide (CO2), 5% Temperature: 37.0°C
Subculturing:
Protocol: Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 X 10(5) cells/ml. Interval: Maintain cell density between 1 X 10(5) and 1 X 10(6) viable cells/ml. Medium Renewal: Every 2 to 3 days
Preservation:
Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO Storage temperature: liquid nitrogen vapor phase
