    MMP-9 是一种糖化前酶。酶的活化涉及到脱 N 端 PRO 区的蛋白水
    解,产生 82kDa 的活性酶。活性人 MMP-9 与小鼠和大鼠 MMP-9 的氨基
    酸序列同源性分别为 72%和 74%。除锌结合位点外,催化结构域还包含
    三个连续的纤维连接蛋白 II 型同源性单元,负责结合明胶。一个富含脯
    氨酸的铰链区将催化结构域连接到 C-端血红素样结构域。用 4-氨*苯
    基乙*汞(APMA)体外处理前酶,不仅产生活性酶,而且产生 C 末端截短
    形式,其活性与活性形式相当。MMP-9 降解 ECM 中对变性胶原具有高
    比活性的成分。它能切割 III 型、IV 型、V 型和 XI 型的天然胶原,以及
    弹性蛋白、尼达根-1 和维托菌素。MMP-9 还能切割多种趋化因子和生
    长因子、淀粉样α肽、P 物质和髓鞘碱性蛋白。这种作用可以增加或降
    低可溶性因子的生物活性,也可以使它们脱离与 ECM 的联系。MMP-9
    还可通过多种跨膜蛋白触发信号传递,或通过诱导其从细胞表面脱落(如
    CD44、E-钙粘附素、整合素、ICAM-1 和 IL-2 Rα)而抑制信号传递。
