最初，用0.25%胰酶消化液传代，细胞脱壁容易，但是，吹打细胞180次左右，也只有80％细胞是单个细胞，而且，易引起细胞机械性损伤。于是，我改进方法。 现在，我的处理如下： 弃旧培养液 0.02％EDTA作用2分钟 弃EDTA 0.1％胰酶作用30秒 弃胰酶，继续消化2分钟 加培养液终止消化。一般吹打细胞30－40次，细胞基本为单个。 EDTA是一种化学螯合剂，主要作用主任已说。我要提醒一下：EDTA作用不受血清抑制，故消化后需彻底清除，否则影响细胞生长。 建议：可能的话离心
