In Vitro
Cell（Chondrocytes；2.5~ 40 μM HSYA；24h）
For the in vitro study, to assess the anti-inflammatory effects of different concentrations of HSYA, cells were treated with 10 ng mL−1 IL-1β alone or in combination with HSYA pretreatment at different concentrations (2.5, 10, or 40 μM). A control group was left untreated except for medium changes. Cells were harvested after 24 h of incubation. To investigate the activation of NF-κB and MAPKs, the duration of IL-1β stimulation was 2 h. Meanwhile, to explore functional changes, such as inflammatory or ECM markers, the duration was extended to 24 h.

In Vivo
Mice（ten-week-old C57BL/6 male wild-type (WT) mice; 20 mg/kg/day HSYA; 腹腔注射，1次/天，8周）
After DMM, the HSYA treatment groups received HSYA administration for 20 mg/kg/day (the HSYA was dissolved in saline and injected intraperitoneally once a day for eight consecutive weeks). Meanwhile, the mice in the DMM alone group received an equivalent volume of saline. All animals were sacrificed after eight weeks post-surgery, and cartilage tissue samples were collected for iconographic and histological analysis.
