    TNF-α 是 TNF 蛋白超家族的原型成员，是一种同源三聚体 II 型膜
    蛋白。膜结合的 TNF-α 被金属蛋白酶 TACE/ADAM17 切割生成可溶性
    同三聚体。TNF-α的膜形式和可溶性形式都具有生物活性。细胞对 TNF-α
    的反应通过与受体 TNF-R1 和 TNF-R2 的相互作用来介导，并根据细胞
    类型和生物学背景激活有利于细胞存活和凋亡的途径。激酶途径（包括
    JNK、ERK（p44/42）、p38 MAPK 和 NF-κB）的激活促进细胞的存活，
    而 TNF-α 介导的半胱天冬酶-8 激活导致细胞程序性死亡。TNF-α 在炎
    症和宿主抵抗细菌感染，尤其是结核*杆菌感染中起着关键的调节作
    用。TNF-α 引起许多与自身免疫性疾病相关的临床问题，如类风湿性关
    节炎、强直性脊柱炎、炎症性肠病、银屑病、化脓性汗腺炎和难治性哮
    喘。TNF-α 在自身免疫中的作用通过阻断 TNF-α 治疗类风湿性关节炎和
    克罗恩病而得到强调。
