    人血小板生成素(TPO)又称巨核细胞生长发育因子(Mgdf)、MPL 配
    体和巨核细胞集落刺激因子(MK-CSF)，是一种 60~70 kDA，332 个氨基
    酸残基糖基化多肽，在巨核细胞的发育过程中起着关键作用。TPO 分子
    主要由肝细胞和骨髓基质细胞分泌，在结构和功能上可分为 n 端和 c 端
    两部分，TPO 的 n 端半部结构类似于促红细胞生成素(EPO)分子，约有
    25%的氨基酸残基同源性和大量半胱氨酸的保守性。这一区域在功能上
    被认为与受体结合有关，而 c-末端区域的结构特征是丝氨酸和脯氨酸残
    基的比例很高，含有许多潜在的 n-连接糖基化位点，c-末端被认为有助
    于整个分子的稳定性和循环半衰期。不同物种的氨基酸残基均有明显的
    保守性，人 TPO 与大鼠、小鼠和狗 TPO 的同源性分别为 75%、73%和
    76%。氨基酸结构的保存是人类和小鼠细胞上发现的跨物种生物活性的
    原因。TPO 能特异性的刺激巨核系祖细胞增殖分化，促进巨核细胞成熟，
    还能强烈促进核细胞或巨噬细胞和红系祖细胞的恢复，有诱导造血祖细
    胞动员进入外周循环的作用。TPO 主要产生于肺和肾脏，血清游离 TPO
    浓度是通过黏附因子表达在人外周血血小板和巨核细胞上的 c-mpl 来
    调节。
