    背景介绍
    IL-28A 也称为干扰素λ2，是 IL-10 家族和 I 型 IFN 家族成员的远亲。
    IL-28A 基因定位于 19 号染色体上，每个基因由多个外显子组成。这些
    基因的外显子组织也在 IL-10 家族基因中发现，但是它不同于 I 型干扰
    素，它只有一个外显子编码。 IL-28A 通过病毒感染或者双链 RNA 诱
    导表达。IL-28A 细胞因子重叠 I 型干扰素发挥生物活性，包括抗病毒活
    性和 MHC I 类抗原表达。IL-28A 蛋白是通过相同的异质二聚体受体复
    合物发射信号，这些受体复合物由 IL-10 受体β和一种新型 IL-28 受体α。
    人的 IL-28A cDNA 编码一个 200 氨基酸（aa）残基前体蛋白和一个一般
    认定的 25 氨基酸信号肽。它与人的 IL-28B 和 IL-29 分别有 94%和 67%
    的氨基酸同源性。
     
    检测原理
    Solarbio（Solarbio ®）ELISA 试剂盒采用基于双抗体夹心法的酶联
    免疫吸附检测技术。将抗人 IL-28A/IFN-λ2 单克隆抗体包被在酶标板上；
    分别加入梯度稀释的标准品和预稀释的样本，标准品和样本中的人
    IL-28A/IFN-λ2 会与酶标板上的包被抗体充分结合；洗板后加入生物素
    化抗人 IL-28A/IFN-λ2 抗体，该抗体会与板子上包被抗体捕获的标准品
    和样本中的人 IL-28A/IFN-λ2 发生特异性结合；洗板后加入辣根过氧化
    物酶（HRP）标记的链霉亲和素，生物素与链霉亲和素会发生高强度的
    非共价结合；洗板后加入显色剂底物 TMB，若反应孔中样品存在不同
    浓度的人 IL-28A/IFN-λ2，则 HRP 会使无色 TMB 变成不同深浅（正相
    关）的蓝色物质，加入终止液后反应孔会变成黄色；最后，在λmax=450
    nm（OD=450 nm）处测定反应孔样品吸光度（OD），样本中的人
    IL-28A/IFN-λ2 浓度与 OD 成正比，通过绘制标准曲线和四参数拟合软
    件便可计算出样本中人 IL-28A/IFN-λ2 的浓度。
