    背景介绍
    IL-12P40 是由二硫键相连异源二聚体,70kDa 的(p70)异二聚体糖蛋
    白由 40kDa(p40)和 35kDa(p35)两个亚基组成。p40 和 p35 亚基没有
    IL-12P40 的活性,只有异二聚体 IL-12P40p70 才具有 IL-12P40 生物学活
    性,IL-12P40p40 同型二聚体可与 IL-12P40 的受体结合,是 IL-12P40 的拮
    抗剂。人 IL-12P40 P35 亚单位有 197 个氨基酸残基,含 7 个半胱氨酸和 3
    个 N 糖基化位点,P40 亚单位有 306 个氨基酸残基,含 10 个半胱氨酸和 4
    个 N 糖基化位点。人类 p40 和 p35 分别在染色体 5 和 3 上。P40 与 IL-6
    受体、睫状神经营养因子(CNTF)受体、G-CSF 受体有同源性,具有细胞
    因子受体家族的特征。
    IL-12P40 也被称为自然杀伤细胞刺激因子(NKSF)
    或细胞毒性淋巴细胞成熟因子(CLMF),是由抗原提成细胞(单核/巨噬细
    胞、树突状细胞核 B 细胞)产的多效细胞因子,可作用于 T 细胞和 NK 细
    胞,促进其增殖、细胞毒性、细胞因子产生和 Th1 亚群的发育。
