质粒简介
pET-21a质粒是一个大肠杆菌表达载体,T7启动子驱动T7标签、6×HIS标签和目的蛋白融合表达。The pET-21a-d(+) vectors carry an N-terminal T7•Tag® sequence plus an optional C-terminal His•Tag® sequence. These vectors differ from pET-24a-d(+) only by their selectable marker (ampicillin vs. kanamycin resistance). Unique sites are shown on the circle map. Note that the sequence is numbered by the pBR322 convention, so the T7 expression region is reversed on the circular map. The cloning/expression region of the coding strand transcribed by T7 RNA polymerase is shown below. The f1 origin is oriented so that infection with helper phage will produce virions containing single-stranded DNA that corresponds to the coding strand. Therefore, single-stranded sequencing should be performed using the T7 terminator primer.

文献和实验晚上每天都有人做报告,我觉得让我收获最大的是Bill Studier的报告。本来这个报告是后来才听到的,但是由于Burgess的模块是唯一的涉及大肠杆菌中表达蛋白的,我把这一段提前来说说。Studier这老哥是Brookhaven National Laboratory的,一生研究T7噬菌体,也是T7 RNA polymerase induction system(pET系列质粒)的发明者。 T7系统在大肠杆菌表达蛋白的应用实在太广泛了,但凡表达蛋白的兄弟姐妹们的不可能不知道这个系统

其分子间作用力呈显著下降趋势。 在计算机模拟和蛋白质分子设计的基础上,以野生型TFPI基因为模板,用PCR方法引入突变,构建表达质粒pET―28a(+)―TFPI―Mutl和pET―28a(+)―TFPI―Mut4。然后参照野生型TFPI的基因表达、蛋白质复性及纯化方式,用大肠杆菌制备重组的TFPI―Mutl和TFPI―Mut4。试验证实TFPI―Mutl和TFPI―Mut4保持了与TFPI基本相当的抗凝活性,而且半衰期较TFPl分别延长1.62和4.22倍。
