Prostaglandin E2(PGE2)是一种具有多种活性的脂质,参与炎性反应和很多肿瘤的形成。目前因其在炎症、癌症、动脉粥样硬化、自身免疫性疾病和脓毒症等疾病调控中的优越表现被广泛研究。花生四烯酸被前列腺素合成酶(COX-1和COX-2)氧化生成前列腺素H2 (PGH2),PGH2进一步被PGE合成酶代谢生成其主要产物PGE2。PGE2通过与细胞表面的g蛋白偶联受体(EP1、EP2、EP3、EP4)结合,介导自分泌和旁分泌信号,调节磷脂酶C和腺苷酸环化酶活性。作为治疗靶点,PGE2一直是科研者研究的热点,无论是在通过COX抑制剂(NSAIDS)对其合成的调节中,还是通过下调或结合拮抗剂对其受体的调节中。各种组织中产生的PGE2已被证明可以调节许多生理过程,包括肾脏的利钠尿、血管平滑肌弹性以及单核细胞和巨噬细胞对受损组织的炎症反应。

PGE2α的生物学意义
Prostaglandin F2α(PGF2α)是由各种细胞中的PGH2形成的, 它本身是由花生四烯酸经前列腺素合成酶合成的。由于PGF2α在各种组织中具有与PGF2相反的作用,因此它通常被认为是PGE2的抑制剂。PGF2α是支气管收缩的激动剂,与哮喘发作有密切关系。由于PGF2α可参与许多生殖器官功能的调节,例如黄体调节、子宫收缩和精子运动等,目前被广泛用于终止妊娠和引产。另外,高水平的PGF2α也与子痫前期相关。
