storage:The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself. Generally, the shelf life of liquid form is 6 months at -20℃,-80℃. The shelf life of lyophilized form is 12 months at -20℃,-80℃.
Notes:Repeated freezing and thawing is not recommended. Store working aliquots at 4℃ for up to one week.
background:Component of the 9-1-1 cell-cycle checkpoint response complex that plays a major role in DNA repair. The 9-1-1 complex is recruited to DNA lesion upon damage by the RAD17-replication factor C (RFC) clamp loader complex. Acts then as a sliding clamp platform on DNA for several proteins involved in long-patch base excision repair (LP-BER). The 9-1-1 complex stimulates DNA polymerase beta (POLB) activity by increasing its affinity for the 3'-OH end of the primer-template and stabilizes POLB to those sites where LP-BER proceeds; endonuclease FEN1 cleavage activity on substrates with double, nick, or gap flaps of distinct sequences and lengths; and DNA ligase I (LIG1) on long-patch base excision repair substrates. The 9-1-1 complex is necessary for the recruitment of RHNO1 to sites of double-stranded breaks (DSB) occurring during the S phase. Isoform 1 possesses 3'->5' double stranded DNA exonuclease activity.

文献和实验Purification and Identification of Protein Complexes That Control the Cell Cycle
The human Rad9-Hus1-Rad1 (9-1-1) and Rad17-RFC40-RFC38-RFC37-RFC36 (hRad17-RFC) protein complexes are important components of the mammalian deoxyribonucleic acid (DNA) damage response. Accumulating evidence from biochemical and molecular
Analyzing Checkpoint Controls in Human Skin
A short-term whole-skin organ culture model has been established to enable the investigation of cell cycle perturbations in epidermal layer cells following exposure to ultraviolet radiation (UVR). This model affords the opportunity to manipulate
Assaying Cell Cycle Checkpoints: Activity of the Protein Kinase Chk1
Eukaryotic cells regulate progression through the cell cycle in response to DNA damage. Cell cycle checkpoints are the signal transduction pathways that couple the detection of DNA damage to the proteins that control transitions in the cell
