LS-F35370 is a 96-well enzyme-linked immunosorbent assay (ELISA) for the Quantitative detection of Human FKBP5 / FKBP51 in samples of Plasma, Serum and Tissue Homogenates. It is based upon a Sandwich assay principle and can be used to detect levels of FKBP5 / FKBP51 as low as 0.094 nanograms per millilter.

Background
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- FKBP5antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- FKBP5 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP- Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the FKBP5 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of FKBP5 can be calculated.

FKBP5 / FKBP51 is a member of the immunophilin protein family, which play a role in immunoregulation and basic cellular processes involving protein folding and trafficking. This encoded protein is a cis-trans prolyl isomerase that binds to the immunosuppressants FK506 and rapamycin. It is thought to mediate calcineurin inhibition.
