    E3 Ubiquitin-Protein Ligase CHIP is a cytoplasmic protein. CHIP is highly expressed in skeletal muscle, heart, pancreas, brain and placenta. CHIP interacts with the molecular chaperones Hsc70-Hsp70 and Hsp90 through its TPR domain; lead to in client substrate ubiquitylation and degradation by the proteasome. CHIP targets misfolded chaperone substrates towards proteasomal degradation. CHIP mediates transfer of non-canonical short ubiquitin chains to HSPA8 that have no effect on HSPA8 degradation. CHIP plays a role in base-excision repair: catalyzes polyubiquitination by amplifying the HUWE1/ARF-BP1-dependent monoubiquitination and leading to POLB-degradation by the proteasome. It also may regulate the receptor stability and activity through proteasomal degradation.

    Transforming Growth Factor α (TGF-α) belongs to the EGF family of cytokines. It is a mitogenic polypeptide and secreted protein, which is expressed by monocytes, keratinocytes, and various tumor cells. TGFα contains two chains, protransforming growth factor α and transforming growth factor α. It can bind to the EGF receptor that synergistically with TGFβ to stimulate anchorage-independent cell proliferation and produce a mitogenic response. TGFα interacts with the PDZ domains of MAGI3, SDCBP and SNTA1. The interaction with SDCBP is required for the targeting to the cell surface.

MAB1B1 is a single-pass type II membrane protein and widely expressed in many tissues. MAB1B1 is involved in glycoprotein quality control targeting of misfolded glycoproteins for degradation.     Amiloride-sensitive amine oxidase (AOC1) belongs to the copper/topaquinone oxidase family. The protein exists as homodimer by disulfide and mainly located in placenta and kidney. AOC1 catalyzes the degradation of compounds such as putrescine, histamine, spermine, and spermidine, substances involved in allergic and immune responses, cell proliferation, tissue differentiation, tumor formation, and possibly apoptosis. Placental DAO is thought to play a role in the regulation of the female reproductive function. The activity of this protein can be inhibited by amiloride in a competitive manner. It is inhibited by amiloride, a diuretic that acts by closing epithelial sodium ion channels.

    Receptors for the Fc region of IgG (FcγR) are members of the Ig superfamily that function in the activation or inhibition of immune responses. Human FcγRs are divided into three classes designated FcγRI (CD64), FcγRII (CD32), and FcγRIII (CD16), which generate multiple isoforms, are recognized. The activating­ type receptor either has or associates non­covalently with an accessory subunit that has an immunoreceptor tyrosine­based activation motif (ITAM) in its cytoplasmic domain. FcγRI binds IgG with high affinity and functions during early immune responses, whereas FcγRII and RIII are low affinity receptors that recognize IgG as aggregates surrounding multivalent antigens during late immune responses. Three genes for human FcγRII (A, B, and C) and one for mouse (FcγRIIB), encoding type I transmembrane proteins with ITAM motifs (FcγRII A and C) or ITIM motifs (FcγRIIB) in their cytoplasmic domains, have been identified. Human CD32, also known as Low affinity immunoglobulin γ Fc region receptor II-a (IgG Fc receptor II-a), FcγRII A or FCGR2A Protein, is expressed on cells of both myeloid and lymphoid lineages as well as on cells of non-hematopoietic origin. Associated with an ITAM-bearing adapter subunit, FcRγ, CD32a (FcγRII A) delivers an activating signal upon ligand binding, and results in the initiation of inflammatory responses including cytolysis, phagocytosis, degranulation, and cytokine production. The responses can be modulated by signals from the co-expressed inhibitory receptors such as Fcγ RII B, and the strength of the signal is dependent on the ratio of expression of the activating and inhibitory receptors.
