使用不同逆转录酶在50℃反应30分钟，将100 ng至1 pg小鼠肝脏总RNA 逆转录为cDNA。取1/10 cDNA为模板，用AceTaq® DNA Polymerase扩增β-Actin基因。NTC: no template control.
HiScript® Reverse Transcriptase是在M-MLV (RNase H-) Reverse Transcriptase基础上经过多点突变得到的全新逆转录酶。HiScript® Reverse Transcriptase在50℃下活性最高，且可耐受高达55℃的反应温度，可用于具有二级结构的RNA模板的逆转录。增强与模板的亲和力，可获得更多全长的cDNA，并可检测到低至1 pg的总RNA，特别适合少量模板以及低拷贝基因的逆转录。此外HiScript® Reverse Transcriptase的错配率也比M-MLV (RNase H-) Reverse Transcriptase有所降低，提高了克隆的保真度。
