Uracil Excision Kit使用的Uracil Excision Enzyme可以特异性地切除尿嘧啶并形成一个单核苷酸缺口。Uracil Excision Enzyme包含有两种酶：尿嘧啶-DNA糖基化酶(UDG)和内切酶VIII(Endo VIII)。UDG催化从含尿嘧啶的DNA上释放尿嘧啶，产生一个脱碱基位点。Endo VIII会裂解这个脱碱基位点处的磷酸二酯键而形成一个单核苷酸缺口。本产品既可以切割单链DNA，也可以切割双链DNA。
