Place on a plate shaker at 300-700 rpm at room temperature (18°C to 28°C) for 30 minutes
Wash 3 times with 1X Wash Buffer
Add 100 µL of the TMB Substrate
With appropriate cover to avoid light exposure, place the microplate(s) on a plate shaker at 300-700 rpm at room temperature (18°C to 28°C) for 15 minutes
Add 100 µL of Stop Solution and read at 450 nm

Vitamin D is a group of fat-soluble secosteroids which are produced in the skin of vertebrates after exposure to ultraviolet light from the sun, available from artificial sources, or occur naturally in a small range of foods. Total 25-OH Vitamin D is a combined measurement of two major forms, vitamin D2(ergocalciferol) and vitamin D3 (cholecalciferol); known collectively as calciferol. When synthesized in the kidneys, calciferol circulates as a hormone; regulating, among other things, the concentration of calcium and phosphate in the bloodstream, promoting the healthy mineralization, growth and remodeling of bone.
