Azido Palmitic Acid (also sold as Click-iT® palmitic acid, azide) can be used to identify and characterize post-translationally palmitylated proteins with using a simple and robust two-step labeling and detection technique. Step 1 – the Azido Palmitic Acid is fed to cells or animals and actively incorporated into proteins. Step 2- the incorporated Azido Palmitic Acid is detected through the chemoselective ligation or “click” reaction between and the incorporated azide group and an alkyne detection reagent. A wide variety of detection reagents are available from Click Chemistry Tools, including fluorescent dyes, hapten or biotin-free enrichment kits.
