Product Description

LS-F39437 is a 96-well enzyme-linked immunosorbent assay (ELISA) for the Quantitative detection of Rat BrdU in samples of Plasma, Serum and Tissue Homogenates. It is based upon a Competitive EIA assay principle and can be used to detect levels of BrdU as low as 0.938 picomoles per milliliter.
About BrdU
Bromodeoxyuridine (5-bromo-2'-deoxyuridine, BrdU, BUdR, BrdUrd) is a synthetic nucleoside that is an analog of thymidine. BrdU is commonly used in the detection of proliferating cells in living tissues. 5-Bromodeoxycytidine is deaminated to form BrdU. BrdU can be incorporated into the newly synthesized DNA of replicating cells (during the S phase of the cell cycle during which DNA is replicated), substituting for thymidine during DNA replication.

Background
 
This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with BrdU. During the reaction,BrdU in the sample or standard competes with a fixed amount ofBrdU on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to BrdU. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin(SABC) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of BrdU in the samples is then determined by comparing the O.D. of the samples to the standard curve.
