产品描述:
热敏UDG（尿嘧啶-DNA 糖基化酶）能有效地水解单链或双链 DNA上的尿嘧啶，产生的缺嘧啶位点，在高温或高pH下，极易水解断裂。热敏UDG酶与常规UDG酶相比，可在较低温度（20℃）下起作用，且对温度敏感、易于灭活，避免了常规UDG酶灭活后可能存在的残留活性在常温下对含dU扩增产物的降解作用。升高温度和pH都会影响其水解活性。该产品对高温敏感，50℃以上就可以将酶完全失活。
产品用途：
1）去除PCR残存污染；
2）去除单链或双链DNA尿嘧啶碱基，5~10min降解含dU的DNA模板；
3）适用于30℃反应，50℃灭活，尤其适合反转录一步法防污染扩增。
使用建议：
热敏UDG在多数PCR反应缓冲液中均有活性，但活性受高离子强度（＞100 mM）抑制；
1×UDG Reaction Buffer条件下，30℃ 温育5-10min；
热失活：50℃加热10min或者95℃ 加热2min。
保存温度：
-20℃
质量保证：
经多次柱纯化，SDS-PAGE胶检测仅可见清晰单一的目的条带，PCR方法检测无大肠杆菌DNA残留，无核酸内、外切酶污染。
活性定义：
1 单位指每分钟催化60pmol尿嘧啶从含尿嘧啶的双链DNA上释放所需要的酶量。检测条件为：50μl标准Taq反应缓冲液中，37℃条件下，30分钟从含0.2μg DNA（104-105 cpm/μg）的反应体系中释放[3H]-尿嘧啶的量来确定活性。
