关于IGF1(Long-R3-IGF1)：
Insulin-like growth factor I (IGF1) belongs to the family of insulin-like growth factors that are structurally homologous to proinsulin. Mature IGFs are generated by proteolytic processing of inactive precursor proteins, which contains the N- and C-terminal propeptide regions. Mature human IGF-I consisting of 70 amino acids has 94% identity with mouse IGF-I and exhibits cross-species activity. IGF-1 binds IGF-IR, IGF-IIR, and the insulin receptor and plays a key role in cell cycle progression, cell proliferation and tumor progression. IGF-1 expression is regulated by growth hormone. R3 IGF-1 is an 83 amino acid analog of IGF-1 comprising the complete human IGF-1 sequence with the substitution of an Arg (R) for the Glu(E) at position three, hence R3, and a 13 amino acid extension peptide at the N terminus. R3 IGF-1 has been produced with the purpose of increasing biological activity. R3 IGF-1 is significantly more potent than human IGF-I in vitro.

  本蛋白酶是通过通过酵母编码的ΜlP1基因，在大肠杆菌中表达，经多次纯化获得。SUMO蛋白酶能够识别并且高效地将SUMO（Small Ubiquitin-Like Modifier）从融合蛋白上切割下来。相对于EK和TEV等蛋白酶的短小识别位点，SUMO蛋白酶能够识别完整的SUMO序列，并依赖于正确的蛋白质构象，所以SUMO蛋白酶不会错切融合的蛋白。SUMO蛋白酶的效力可维持在较宽范围的反应环境体系中，例如温度（4～30℃）、pH（5.5～9.5）等。SUMO蛋白酶还具有多聚His标签，便于融合蛋白切割后的亲和层析纯化。本SUMO蛋白酶的浓度为5000U/ml


图例一）SUMO蛋白酶酶切反应：每个泳道含有15μg的目标蛋白与20U/mg比例进行SUMO酶切的上样。在室温（25℃）经过不同时间反应后用15%的SDS-PAGE胶检测酶切效果。
每个反应所用酶切时间分别为：
Lane 1: No Sumo蛋白酶
Lane 2: 5 units/mg
Lane 3: 10 units/mg
Lane 4: 20 units/mg
Lane 5:蛋白Marker


使用建议：因为不同蛋白底物，SUMO蛋白酶的酶切效率会有差异，可以在大量酶切前，先小量酶切测试在不同温度下的酶切效率。

活性定义：在1×SUMO Protease Buffer（无盐）反应体系中(50mM Tris-HCl，pH 8.0，0.2% Igepal，1mM DTT)，加入1U SUMO Protease及5μg的反应底物（His-SUMO-GFP）补H2O至总体积100μl，25℃条件下反应1小时，酶切效率达90%以上。

质量保证：经多次柱纯化，SDS-PAGE胶检测仅可见清晰单一的目的条带；本制品不含有非特异性的蛋白酶切活性。

储存条件：-20℃，避免反复冻融，有效期12个月。

关于Complement C5：
Mouse Complement C5 (C5a) is a glycoprotein that belongs to a family of structurally and functionally related proteins known as anaphylatoxins. C5a is a 77 amino acid peptide that is created by the C5a convertase proteolytic cleavage of C5 αchain in the classical and alternative complement pathway (C4b2a3b, C3bBb3b). Mouse C5a has fourαhelices, plus three intra-chain disulfide bonds that form a triple loop structure. C5a functions via G-protein coupled receptor (GPCR) (C5aR/CD88). C5a is a potent chemoattractant and anaphylatoxin that acts on all classes of leukocytes and on many other cell types including endothelial, smooth muscle, kidney, liver, and neural cells. It mediates IL-8 release from bronchial epithelial cells. It also triggers an oxidative burst in macrophages and neutrophils, causing release of histamine in basophils and mast cells. C5a anaphylatoxin activity on hepatocytes results indirectly from interaction with nonparenchymal cell via prostanoid secretion. Mouse C5a shares 60% and 82% sequence identity to human and rat C5a, respectively.
