Background	Damage to the Bcl-2 gene has been identified as a cause of a number of cancers, including melanoma, breast, prostate, chronic lymphocytic leukemia, and lung cancer, and a possible cause of schizophrenia and autoimmunity. It is also a cause of resistance to cancer treatments. Antibodies to Bcl-2 can be used with immunohistochemistry to identify cells containing the antigen. In healthy tissue, these antibodies will react with B-cells in the mantle zone, as well as some T-cells. However, there is a considerable increase in positive cells in follicular lymphoma, as well as many other forms of cancer. In some cases, the presence or absence of Bcl-2 staining in biopsies may be significant for the patient"s prognosis or likelihood of relapse.

Background	The formation of the spliceosome includes the assembly of Sm proteins inan ordered manner onto snRNAs. This process is mediated by the survivalof a motor neuron (SMN) protein, and is enhanced by modification of specificArginine residues in the Sm proteins to symmetrical dimethylarginines(sDMAs). sDMA modification of Sm proteins is catalyzed by the methylosome,a complex comprised of the type II methyltransferase PRMT5 (alsodesignated JAK-binding protein 1, JBP1), pICln and two novel factors. PRMT5binds the Sm proteins via their Arginine- and Glycine-rich (RG) domains, whilepICln binds the Sm domains. PRMT5 is a distinct member of the protein-Arginine methyltransferase (PRMT) family, and predominantly localizes tothe cytoplasm in a wide variety of tissues. PRMT5 also associates specificallywith the transcription start site region of the cyclin E1 promoter and, therefore,is involved in the control of transcription and proliferation.The geneencoding human PRMT5 maps to chromosome 14q11.2.

Background	GPX1 is ubiquitously expressed in many tissues, where it protects cells from oxidative stress. Within cells, it localizes to the cytoplasm and mitochondria. As a glutathione peroxidase, GPx1 functions in the detoxification of hydrogen peroxide, specifically by catalyzing the reduction of hydrogen peroxide to water. The glutathione peroxidase also catalyzes the reduction of other organic hydroperoxides, such as lipid peroxides, to the corresponding alcohols. GPx1 typically uses glutathione (GSH) as the reductant, but when glutathione synthetase (GSS) is, as in brain mitochondria, γ-glutamylcysteine can serve as the reductant instead. The protein encoded by this gene protects from CD95-induced apoptosis in cultured breast cancer cells and inhibits 5-lipoxygenase in blood cells, and its overexpression delays endothelial cell death and increases resistance to toxic challenges, especially oxidative stress. This protein is one of only a few proteins known in higher vertebrates to contain selenocysteine, which occurs at the active site of glutathione peroxidase and is coded by the nonsense (stop) codon TGA.
