关于SLAMF7/CD319：
SLAMF7 is a single-pass type I membrane protein and contains 1 Ig-like C2-type (immunoglobulin-like) domain. SLAMF7 is expressed in NK cells, activated B-cells, NK-cell line but not in promyelocytic, B-cell lines, or T-cell lines. Although the cytoplasmic domain of CS1 contains immunoreceptor tyrosine-based switch motifs (ITSM), which enables to recruite signaling lymphocyte activation molecule (SLAM)-associated protein (SAP/SH2D1A), it activates NK cells in the absence of a functional SAP. SLAMF7 positively regulated natural killer cell functions by a mechanism dependent on the adaptor EAT-2 but not the related adaptor SAP. However, in the absence of EAT-2, CRACC potently inhibited natural killer cell function. It was also inhibitory in T cells, which are typically devoid of EAT-2. Thus, SLAMF7 can exert activating or inhibitory influences on cells of the immune system depending on cellular context and the availability of effector proteins.

Human protein N-terminal glutamine amidohydrolase (WDYHV1) is an enzyme that in humans is encoded by the WDYHV1 gene, belongs to the NTAQ1 family.WDYHV1 mediates the side-chain deamidation of N-terminal glutamine residues to glutamate, which is an important step in N-end rule pathway of protein degradation. Conversion of the resulting N-terminal glutamine to glutamate renders the protein susceptible to arginylation, polyubiquitination and degradation as specified by the N-end rule. However,it does not act on substrates with internal or C-terminal glutamine andnon-glutamine residues in any position. With the exception of proline, all tested second-position residues on substrate peptides do not greatly influence the activity. In contrast, a proline at position 2, virtually abolishes deamidation of N-terminal glutamine.

文献和实验应用流式细胞术（FACS）测定动员后的外周血及采集的自体外周血造血干细胞（APBSC）中的CD34+细胞及其亚群，操作简单、快速、重复性好，对及时掌握最佳采集时机，准确判断采集的干/祖细胞数量具有重要指导意义。为准确检测外周血及APBSC中的CD34+细胞，本文比较了几种FACS检测CD34+细胞方法的异同。1、材料与方法1.1 外周血造血干细胞 APBSC采集自经环磷酰胺（CTX）＋重组人粒细胞集落刺激因子（rhG-CSF）动员后的实体瘤患者（淋巴瘤、乳腺癌），采集使用CS3000
x 106/ml； 3.4 加入1 000 U/ml 的重组人IFN-g培养； 3.5 24h 后加入50ng/ml 的CD3 单克隆抗体和300 U/ml 的重组人IL-2，刺激CIK 细胞的生长和增殖； 注：此时也可同时加入100 U/ml的重组人IL-1a。 3.6 每3天半量换液或扩瓶一次，并补加重组人IL-2 300 U/ml； 3.7 在培养的第7d，收获CIK细胞，此时数量应达到1x 109个以上。 3.8 CIK细胞质控： 3.8.1 台盼蓝染色检测
的培养及鉴定 2.1 将PBMC按1-2 x 106/ml的浓度悬浮于无血清培养液中，加入1,000 U/ml 的重组人IFN-g，37oC，5%CO2培养箱中培养； 2.2 24h 后加入50ng/ml 的CD3 单克隆抗体和300 U/ml 的重组人IL-2，刺激CIK 细胞的生长和增殖； 注：此时也可同时加入100 U/ml的重组人IL-1a。 2.3 每3天半量换液或扩瓶一次，并补加重组人IL-2 300 U/ml； 2.4 在培养的第14d，收获CIK细胞。
