关于DNA Polymeraseβ：
Human DNA polymerase β is constitutively expressed in cells. It fills in gaps in DNA that are formed following base excision repair. Repair polymerase that plays a key role in base-excision repair. Has 5-deoxyribose-5-phosphate lyase (dRP lyase) activity that removes the 5 sugar phosphate and also acts as a DNA polymerase that adds one nucleotide to the 3 end of the arising single-nucleotide gap. It conducts gap-filling DNA synthesis in a stepwise distributive fashion rather than in a processive fashion as for other DNA polymerases. The activity cannot be affected by Aphidicolin, which is an inhibitor of DNA polymerase β.

Parafibromin is expressed in the adrenal and parathyroid glands, kidney, and heart. As a tumor supressor, Parafibromin may be involved in transcriptional and post-transcriptional control pathways, also through the regulation of cyclin D1/PRAD1 expression, involved the cell cycle progression. Parafibromin is a component of the the PAF protein complex and interacts with a Set1-like complex that has histone methyltransferase activity and methylates histone H3. Defects in Parafibromin can cause hyperparathyroidism-jaw tumor syndrome, familial isolated hyperparathyroidism, and parathyroid carcinoma.

关于CD33/Siglec-3：
CD33 is a type I Lectin belonging to the Ig superfamily. CD33 contains an N terminal Ig like V type domain, which mediates sialic acid binding, followed by one Ig like C2 type domain, a transmembrane region and a cytoplasmic tail containing two conserved immunoreceptor tyrosine based inhibition motifs (ITIMs). Eleven human Siglecs have been characterized. Siglecs 5 to 11 share a high degree of sequence similarity with CD33/Siglec3 both in their extracellular and intracellular regions. They are collectively referred to as CD33 related Siglecs. CD33 related Siglecs have differential expression pattern within the hematopoietic system. They are involved in the regulation of cellular activation within the immune system. Siglec 3 expression is restricted to cells of myelomonocytic lineage. Siglec3 recruits SHP1 and SHP2 to its ITIMs upon phosphorylation.

Growth/differentiation factor 8(Mstn, GDF-8) is a member of the bone morphogenetic protein (BMP) family and the TGF-beta superfamily. This group of proteins is characterized by a polybasic proteolytic processing site which is cleaved to produce a mature protein containing seven conserved cysteine residues. It is expressed specifically in developing and adult skeletal muscle. It exists as a homodimer, and interacts with WFIKKN2, leading to inhibit its activity. This protein can act specifically as a negative regulator of skeletal muscle growth. It regulates cell growth and differentiation in both embryonic and adult tissues.
