Mannoside Acetylglucosaminyltransferase 2 (MGAT2) is a single-pass type II membrane protein that contains the typical glycosyltransferase domains: a short N-terminal cytoplasmic domain, a hydrophobic non-cleavable signal-anchor domain and a C-terminal catalytic domain. MGAT2 catalyzes an essential step in the conversion of oligo-mannose to complex N-glycans. Defects in MGAT2 are the cause of congenital disorder of glycosylation type 2A.

关于Galectin-8：
The Galectin family of proteins, with specificity for Nacetyllactosaminecontaining glycoproteins, consists of beta-galactoside binding lectins containing homologous carbohydrate recognition domains (CRDs). They also possess hemagglutination activity, which is attributable to their bivalent carbohydrate binding properties. Galectins are active both intracellularly and extracellularly. Although they are localized primarily in the cytoplasm and lack a classical signal peptide, galectins can also be secreted by one or more unidentified, non-classical, secretory pathways. They have diverse effects on many cellular functions including adhesion, migration, polarity, chemotaxis, proliferation, apoptosis, and differentiation. Galectins may therefore play a key role in many pathological states, including autoimmune diseases, allergic reactions, inflammation, tumor cell metastasis, atherosclerosis, and diabetic complications. The galectins have been classified into the prototype galectins(1, 2, 5, 7, 10, 11, 13, 14), which contain one CRD and exist either as a monomer or a noncovalent homodimer. The chimera galectins(Galectin3) containing one CRD linked to a nonlectin domain, and the tandemrepeat Galectins(4, 6, 8, 9, 12) consisting of two CRDs joined by a linker peptide.Galectins lack a classical signal peptide and can be localized to the cytosolic comp*(代"a")rtments where they have intracellular functions. However, via one or more as yet unidentified nonclassical secretory pathways, galectins can also be secreted to function extracellularly.   本试剂盒采用温和的溶胶液，可以使琼脂糖凝胶完全融化，同时避免在高温下DNA发生脱嘌呤和末端水解，保持DNA完整的生物学活性。采用硅胶膜选择性吸附DNA的方法回收DNA，适合从TAE或者TBE琼脂糖凝胶中纯化回收多至4μg，长度介于50bp-10kb的DNA片段，回收率为90%.。纯化回收的DNA可直接酶切、连接、转化细菌、测序、PCR、杂交等后续操作。

注意事项：
1、Wash Buffer在使用前按照试剂瓶所示体积加入无水乙醇，混合均匀。
2、Buffer GL在较低温度下易沉淀，若出现沉淀，使用前请在37℃加热几分钟，至沉淀溶解后再使用。
3、建议使用TAE电泳分离DNA，因为TBE中的硼*(代"酸")与琼脂糖生成非共价结合的四羟基硼*(代"酸")盐复合物而降低DNA回收率。
4、尽量减少紫外照射的时间，以免损伤DNA。
5、水浴期间要间断性摇晃离心管，加速溶胶进行下一个步骤，减少DNA回收率的降低。

操作步骤：
1、将切割的凝胶转入预先称重的1.5ml离心管中，加入等体积的Buffer GL(100mg的凝胶加入100μl Buffer I)。放入55-60℃水浴中，间断摇晃混合，直至凝胶完全融化（约5-10min），放置使其冷却至室温。
2、将上述混合液加入DNA回收柱内，如果溶液体积>700μl，分次转移溶液；室温放置1-2min或更长时间。
3、在13000g离心1分钟，弃废液。目的DNA长度≤500bp时，离心结束后将收集管中的溶液再次转入DNA纯化柱中，离心。
4、在DNA纯化柱中加入650μl Wash Buffer，13,000g离心30秒，弃废液，将DNA纯化柱放回收集管。
5、重复步骤4。
6、13000g离心3 min，以去除柱中残余乙醇。
7、将纯化柱放入新的离心管中，向柱中加入在60℃下预热的30-50μl Elution Buffer或ddH2O，室温放置1-2min或更长时间。
注意：
a. 加入Elution Buffer后将柱子整个温育5min后再离心洗脱可以增加DNA回收效率。
b. 对大于8Kb的片段，加溶液Elution Buffer后将柱子整个温育15-30min可以提高回收效率。
8、13,000g离心1 min，所得液体即为高纯度DNA。

储存条件：室温，有效期12个月。

CST7 is a secreted protein and primarily expressed in peripheral blood cells and spleen.It is belongs to the cystatin family. The cystatin superfamily encomp*(代"a")sses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens.
