细胞培养:
1、培养前准备
(1)培养器具:推荐使用25 cm2培养瓶或60 mm培养皿(或更小)。
(2)培养基:准备适合培养AcceGen小鼠胚胎成纤维细胞NIH-3T3细胞的培养基,建议用量5 mL(或更少)。
2、细胞解冻
将包装中的冻存管取出后立即放在不高于37℃的水中,并在2分钟内摇晃解冻。
3、细胞复苏
(1)取10ml培养基置于离心管中,用70%酒精或阳离子洗涤剂渗透纱布消毒冻存管,用消毒纱布包裹管身,拧开管盖后,将细胞悬浮液转移到离心管中;
(2)1000 rpm离心3分钟,弃掉上清液;
(3)将细胞重新悬浮在5ml上述培养基中(不清洗细胞)。在25 cm2的培养瓶或60 mm的培养皿中进行培养。
4、细胞传代
在传代之前,确保细胞增殖,因为过度稀释可能导致细胞死亡,尤其是血细胞和淋巴细胞。
5、细胞冻存
待AcceGen小鼠胚胎成纤维细胞NIH-3T3细胞生长状态良好时,尽早进行细胞冻存。

| Description | NIH-3T3 cells, also recognized as Swiss 3T3, trace their origin to a mouse NIH/Swiss embryo and represent a fibroblast cell line characterized by distinctive attributes. When dispersed randomly on a chitosan surface, these cells assume a spindle-like morphology, while alignment on an OCMCS surface induces an elongated form. Despite an initial penchant for extreme contact inhibition, this hallmark has progressively waned within the cell line over successive passages.

文献和实验NIH3T3细胞中本身是没有TK的,1981年Scolnick利用lambda载体把胸腺嘧啶激酶(TK)转入NIH3T3细胞,构建成NIH3T3 TK细胞,所以细胞分为NIH3T3 TK+和NIH3T3 TK-两种细胞,在Scolnick认为NIH3T3就是NIH3T3(TK-)。 他做转染细胞的基础是基于1969年Todaro的实验。 具体内容参看建株文献:J Virol.1981 September; 39(3): 935–944
(1)准备大量的3T3细胞,每个安瓿瓶装1-1000000个细胞,然后冻存。使用的细胞不超过20代。(2)将3T3细胞加入175cm2的培养瓶中,用含有10%小牛血清DMEM培养。案每平方厘米15000个细胞接种。2天后换液,每4-5天传代。(3)通过60Gy X射线或Co饲养细胞灭活。被照射的3T3细胞在4度条件下可存放3-4天。(4)在无照射源的条件下,用丝裂菌素C灭活饲养细胞。(5)在37度条件下,用400微克每毫升的丝裂菌素C处理单层3T3细胞1h。(6)用胰蛋白酶消化丝裂菌素C处理
小鼠胚胎成纤维细胞的富集1、给13-14天的孕鼠注射大约0.5ml阿佛丁。当鼠麻醉后,实施断颈法处死小鼠。2、用70%乙醇擦拭腹部,把皮肤向后拉,暴露出腹膜。用消毒过的工具,剪开腹壁以暴露出子宫角。将子宫角移到10cm的皿里。用10ml不含钙镁离子的PBS洗三遍。3、用剪刀剪开每一测的胚囊,并将胚胎移到培养皿中。4、用两副钟表镊子将胎盘和膜与胚胎分离开,分离后切除内脏(所有深色的东西)。将胚胎转移到(有盖)培养皿中,用10ml不含钙镁离子的PBS洗三遍。5、用带有弯钩的眼科剪将组织剪碎,当你剪
