关于Calumenin:
Calumenin is a secreted calcium-binding protein that belongs to the CREC family. Calumenin contains six EF-hand domains and is expressed at high levels in the heart, placenta and skeletal muscle. Human Calumenin is synthesized as a 315 amino acid precursor that contains a 19 amino acid signal sequence, and a 296 amino acid mature chain. Calumenin localizes to the endoplasmic reticulum (ER) and sarcoplasmic reticulum (SR) of mammalian tissues which plays a role in ER functions as protein folding and sorting. Calumenin is involved in the regulation of vitamin K-dependent carboxylation of multiple N-terminal glutamate residues. It seems to inhibit γ-carboxylase GGCX.

关于TNFRSF3:
It is a single-pass type I membrane protein and contains 4 TNFR-Cys repeats. The protein is a member of the tumor necrosis factor (TNF) family of receptors. It is expressed on the surface of most cell types, including cells of epithelial and myeloid lineages, but not on T and B lymphocytes. The protein is the receptor for the heterotrimeric lymphotoxin containing LTA and LTB, and for TNFS14/LIGHT. It promotes apoptosis via TRAF3 and TRAF5 and may play a role in the development of lymphoid organs. The encoded protein and its ligand play a role in the development and organization of lymphoid tissue and transformed cells. Activation of the encoded protein can trigger apoptosis. Not only does the TNFRSF3 help trigger apoptosis, it can lead to the release of the cytokine interleukin 8. Overexpression of TNFRSF3 in Human Cells cells increases IL-8 promoter activity and leads to IL-8 release. TNFRSF3 is also essential for development and organization of the secondary lymphoid organs and chemokine release.

Fibronectin type III domain-containing protein 5, the precursor of irisin, is a protein that is encoded by the FNDC5 gene.Human Irisin is synthesized as a 212 amino acid (aa) precursor encoding a type 1 transmembrane protein with a 121 aa extracellular domain (ECD), a 21 aa transmembrane domain, and a 39 aa cytoplasmic domain. The ECD of Irisin contains a fibronectin type III domain and multiple glycosylation sites. The ECD is proteolytically cleaved to release the 112 aa soluble Irisin hormone into circulation.Mature human, mouse share 100% sequence identity.Irisin induces expression of peroxisome proliferatoractivated receptor γ coactivator 1α (PGC1α) and uncoupling protein1(UCP1), mitochondrialassociated metabolic proteins. Irisin induces the transition of white adipose tissue into more metabolically active beige adipose tissue.Irisin also regulates neuronal cell differentiation and neurite outgrowth in the brain and is involved in the differentiation of osteoblasts.

The Glypicans are a small multigene family of GPI-linked proteoglycans that play a key role in growth factor signaling. Human Glypican 1 (GPC1) is synthesized as a 558 amino acid (aa) preproprecursor that contains a 23 aa signal sequence, a 507 aa mature segment, and a 28 aa C-terminal prosegment. There are two potential N-linked and four potential O-linked sites for glycosylation or glycanation. There are potentially two heparan sulfate (HS) modifications on GPC1 that could contribute to a native molecular weight of approximately 200 kDa. Mature human GPC1 shares 91% aa identity with mature mouse GPC1. Cells known to express GPC1 include neurons, smooth and skeletal muscle cells, keratinocytes, osteoblasts, Schwann cells, immature dendritic cells, and tumor, plus tumorassociated vascular endothelial cells. The function of GPC1 is complex and varied. As a proteoglycan, it appears to make use of its HS adduct to imp*(代"a")ct select growth factor activity. This is accomplished by having juxtramembrane HS attachment sites, and a flexible, GPI-linkage.
