| 背景描述 | Human periodontal fibroblasts were isolated from periodontal tissue. The periodontal membrane (periodontal ligament, periodontal space) is composed of dense connective tissue. Most of the fibers are arranged in bundles, one end of the fibers is buried in the cementum, and the other end is buried in the bone wall of the alveolar fossa, so that the teeth are fixed in the alveolar fossa. There are nerve, blood vessel, lymph and epithelial cells in the periodontal membrane. Fibroblast is the main cellular component of loose connective tissue, derived from embryonic mesenchymal cells. Fibroblasts are large and have clear outline. They are mostly spindle-shaped or star-shaped flat structures with regular oval nuclei and large and obvious nucleolus. Fibroblasts have strong functional activities, weak basophilic cytoplasm, and obvious protein synthesis and secretion activities. Under certain conditions, they can realize mutual transformation with fibroblasts. Fibroblasts play an important role in the repair of various degrees of cell degeneration, necrosis and tissue defect. Periodontal fibroblasts (PLFs), as the main cells of the periodontal membrane, are the main interstitial cells of the periodontal membrane. They not only have the function of synthesating collagen, matrix, elastic fiber and glycoprotein, but also have the ability to absorb collagen and phagocytic foreign bodies. They also participate in the pathological change, repair and regeneration of periodontal tissue. Nowadays, the use of periodontal membrane cells to establish in vitro models has become an important means to study periodontal diseases. |

倒去培养基,先加入PBS1ml,再加入0.25%胰酶1ml,轻轻晃动培养瓶,使之分布均匀;约10秒。将培养瓶迅速转至镜下观察,镜下见细胞触角变钝,细胞变圆,将瓶侧立回到无菌台内,到掉胰酶,加入2mlDMEM培养基(内涵20%胎牛血清),终止消化,弯管吹打成单细胞,可再在镜下观察,确定是否吹打完全。按1:3或1:2传代,加入适量新鲜培养基后,1.5-2.0ml。放入CO2培养箱。注意:时间宁短勿长。宁愿消化不足也不能消化过,PDLC细胞不能等到细胞长融合再传代,长到80%左右就可以传代
为同一类型的细胞,如上皮细胞或成纤维细胞,也仍具有异质性,在分析细胞生物学特性时比较困难。其次,由于供体的个体差异及其他一些原因,细胞群生长效果有时也不一致。原代培养也是建立各种细胞系(株)必经的阶段。假如原代培养能够维持几小时甚至更长,即可进行进一步筛选。有的细胞具有继续增殖能力,有的细胞类型只是存活而不增殖,而另外一些细胞只是在特殊条件下应用而不存活,因而细胞类型的分布将会改变。在单层培养的情况下,瓶底全部铺满细胞,达到会合以后,对密度有依赖性的细胞则逐渐减少生长,而失去密度依赖敏感性的细胞
