| Specificity | Anti-His Tag Antibody (22H1), mAb, Rabbit recognises 6×His epitope tag localized at the C-terminal, N-terminal, and internal region of fusion proteins. It demonstrates high affinity for 10×His, 6×His, and 5×His, with reduced affinity for 4×His and 3×His. Cross-reactivity with yeast lysates is negligible, while slight cross-reactivity is observed with crude E. coli and mammalian lysates. |

| Target Background | The His epitope tag, commonly referred to as the His-tag, is a short sequence of histidine residues—typically 6, but sometimes ranging from 4 to 10—added to proteins for purification and detection purposes. Proteins tagged with His residues can be easily captured by a metal-chelating resin and subsequently eluted under mild conditions, preserving protein activity and structure. His-tags can be positioned at either the N- or C-terminus of the protein, and their small size minimizes potential interference with protein function. In addition to purification, His-tags are also valuable in Western blotting, ELISA, and immunoprecipitation, where anti-His antibodies are used for detection. Due to their versatility and simplicity, His-tags have become a standard tool in recombinant protein production and research. |
