稳定转染细胞株的构建
原理：
稳定转染，就是转染的质粒DNA整合到宿主细胞染色体上，使宿主细胞可长期表达目的基因及蛋白。需要在瞬时转染基础上对靶细胞进行筛选或者应用高转染效率的病毒，根据不同基因载体中所含有的抗性标志选用相应的药物进行细胞传代，从而得到可稳定表达目的基因的细胞系。

观察目的基因及其表达蛋白在某种细胞中的功能（稳定，可长期进行研究）
一般流程：
1） 筛选浓度测定：以10~14天细胞全部死亡的抗生素浓度为筛选浓度
2） 细胞接种：转染实验前天接种细胞，各种细胞的平板密度依据各种细胞的生长率和细胞形状而定。进行转染当天细胞密度应达到60%~80%覆盖
3） 细胞转染（病毒、脂质体、电穿孔、FuGENE 6）
4） 利用质粒上含有的抗性选择进行筛选
5） 鉴定筛选结果

l 待转染的细胞系（1×106个细胞，可传代，倍增时间小于48小时）以及细胞培养条件的说明
